Dumb question, can I safely unplug the JB4's OBD2 cable, prior to turning the car on, so I can run Kia's built in UVO diagnostics? I like to keep my UVO app up to date and it's been a while since I've run the diagnostics and sent mileage, etc. to the UVO app.

I just want to turn the car on with nothing plugged into the OBD2 port, run the diagnostics, turn the car back off, then plug the JB4 OBD2 connector back in. I won't be doing any driving with the OBD2 cable unplugged.

I'm assuming it won't make any difference as long as I don't try driving with the OBD2 cable unplugged. Or does the JB4 do something with that cable at startup?

Unplug it as often as you like for any reason you like. ;)

If you set your jb4 to map 0 when you leave the car, the next time you start it up, the jb4 will be in "disable" mode.....which is basically telling the OBD "nothing is there". UVO should be no problem
